An antibody specific for ACAN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyACAN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for ACAN is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACAN bound in the initial step. The color development is stopped and the intensity of the color is measured. Along with Type-II collagen, aggrecan forms a major structural component of cartilage, particµLarly articµLar cartilage.Aggrecan consists of two globµLar structural domains at the N-terminal end and one globµLar domain at the C-terminal end, separated by a large domain heavily modified with glycosaminoglycans. The two main modifier moieties are themselves arranged into distinct regions, a chondroitin sµLfate and a keratan sµLfate region.
