An antibody specific for DLGAP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDLGAP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for DLGAP1 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DLGAP1 bound in the initial step. The color development is stopped and the intensity of the color is measured. Product Overview:DLGAP1encodes the protein called guanylate kinase-associated protein (GKAP). GKAP binds to the SHANK and PSD-95 proteins fascilitating the assembly of the post synaptic density of neurons. Sequence analysis predicted that the 627-amino acid hydrophilic protein, which is 98% identical to the rat sequence, contains five 14-amino acid repeats in its N terminus. Binding analysis determined that the N-terminal region of DLGAP1 interacts with intact GK domains of all members of the DLG family, which share approximately 75% homology in the GK domain; DLGAP1 was found to interact less well with more distantly related MAGUKs. Immunoblot analysis showed expression of 95- and 130-kD DLGAP1 proteins in membranes from different regions of rat brain. Stability:The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
