An antibody specific for PTGS2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPTGS2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for PTGS2 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTGS2 bound in the initial step. Cytochrome c oxidase (EC 1.9.3.1) is an oligomeric enzymatic complex which is a component of the respiratory chain and is involved in the transfer of electrons from cytochrome c to oxygen. In eukaryotes this enzyme complex is located in the mitochondrial inner membrane; in aerobic prokaryotes it is found in the plasma membrane. The enzyme complex consists of 3-4 subunits (prokaryotes) to up to 13 polypeptides (mammals). In Leigh's disease, there may be an abnormality or deficiency of cytochrome oxidase.
